Alluka is quite the powerful girl
Killua considers himself to be the closest to Alluka, and Killua respects Gotoh a lot compared to other members of the Zoldyck estate. The reason why he corrects Gotoh there is because Killua considers Alluka as his 妹 (younger sister). He wants Gotoh to respect his perspective on Alluka.
Contrary to Western culture, the way you address a person's gender in Japanese is from the addressee's perspective, and not from the person himself/herself. The thing is, Japanese avoid using 3rd person as much as possible to avoid disrespect. So the way the author let us understand how the Zoldyck family rejects and disrespect Alluka is by emphatically referring him/her as a "thing" rather than a person. Killua's way to refer Alluka as his "sister" is only his personal choice. (SPOILER ALERT) A few episodes later, Killua even calls Alluka his 弟 (younger brother) when talking to Morel on the phone. Killua doesn't mind this because his relationship with Morel isn't as close as Gotoh's.
There is in fact no way for the reader to know what gender/sex is Alluka. Anyone who says otherwise only tries to project their view on the subject.

Since English is structured in a way where the subject must always be present in the sentence (Japanese leave out pronouns most of the times, especially 3rd person), translators must make a choice for characters who got an ambiguous gender. Japanese isn't focused on the "ego" as much as English does structurally.
That said, since I have both read and watched Hunter x Hunter in Japanese, I can say for certain that Togashi purposefully left Alluka's gender/sex ambiguous for the reader. From a Japanese thinking perspective, the family emphatically rejects Alluka whenever they address him/her as a "thing". The misgender thing is only a western idea. The gender is, for most Japanese, the perspective of the addressee and not the person himself/herself. In other words, there is no "misgender" as there are no gender clearly specified.
In fact (SPOILER ALERT HERE), a few episodes later when Killua is on the airship talking to Morel, he calls Alluka "弟" (younger brother) instead of "妹" (younger sister). When Illumi wants to provoke Killua, he addresses Alluka as a "thing", not as a brother.

It’s not confirmed her gender is ambiguous. For me she’s a biological girl who is despised by her family and only killua respects her. That’s how I see it. Until Togashi confirms it then we all have accept it until then we interpret her character as we see fit.

Honestly this is very common in anime or manga with androgynous characters. Hange in the manga of AOT there’s no mention of her gender and if I’m not mistaken Hajime want the reader to interpret Hange as they see fit. In the anime she’s confirmed to be a woman but only in the anime. Similar situation happened to Pitou whereas in the manga she has a more androgynous look whereas in the anime she has breast and she’s more feminine and she’s voiced by a woman so the vast majority of the viewers sees her as a female character. It’s worth to mention that it’s ok if people see her as non-binary nothing wrong with it since it wasn’t really confirmed. Though I asume in the anime she’s female. Another good example is Uraume from JJK though with her gender is more ambiguous so here either gender is acceptable. As for Alluka I see her as a biological girl who is despised by her family for being born a girl and for having a weird murderous entity in her. So her family just don’t care about her but Killua. That’s how I see it and makes more sense to me. Again if people see her as a trans girl go for it. I like that you didn’t confirm the fact that she’s ambiguous and nothing is confirmed. Until Togashi confirms it until then we can interpret her character as we see fit.

I would buy it and respect it if I heard directly from Togashi. Has he said this? Or is this an "after the fact" interpretation by 3rd-parties>

This arc is very underrated, mostly because it's so short. But to me, it gives off a lot of Yorknew vibes. Also new interesting characters. Safe to say it now since you already saw her, But Alluka is one of the highlights/best things about this arc. I think you'll also like another character later on.

Not saying this is fact, but I heard this discussed: The family calling Alluka "he" and Killua calling Alluka "she" is a commentary on trans representation. The family rejects Alluka fully. They see her as a monster, something that could wipe out the family if they're not careful. Killua is the only one who saw Alluka as his sister, not something else. From his deep understanding of Alluka, he knows Alluka is a she. The rest of the family rejects this, and calls her "he". Again, I have no idea if this is from Togashi himself, so take it with a grain of salt.

It isn't a mistranslation but you are right that she is the youngest sister

It is not a mistranslation at all. You just don't understand the--honeatly, super obvious--gender dynamics Togashi wrote into the family.

Alluka’s case is really weird lol. I have understood that yes Alluka is indeed a girl but since the rest of the family don’t care about her. They just changer her pronouns from masculine pronouns to it/its pronouns. Killua is the best brother.

I love that you guys immediately picked up on how the family refers to Alluka vs how Killua refers to Alluka and what the meaning behind that could be. I remember being confused about this when I first saw it, thinking it was just errors in the subtitles since Japanese pronouns are vague. But when you listen closer, the butlers also refer to Alluka as a boy ("bocchan"/young son of a rich family) and Killua definitely thinks of her as a girl.
It was nice to see all the cameos but I gotta admit, I was definitely more interested in the Alluka stuff than the Hunter election. Glad to have Hisoka back here to liven things up -- for better or worse, he always adds an extra oomph in any arc he's involved in.

Alluka is Kilua's sister. Just a miss translation on Eng dub. Just a heads up, they mistankenly say brother/he a few times in the upcoming episode
