In order for an alchemist to be able to transmute they need to know what elements the thing they are transmuting is made out of. That was why Ed was all "WTF This thing's made of iron isn't it?" when his transmutation didn't work. His alchemy was not disabled like what Father did in their fight with him.

2023-06-23 15:48:05 +0000 UTCThe reason why Eds alchemy didn’t work on that automail was alluded to, he thought it was made of iron like his and was trying to transmute iron. His alchemy was still working since we saw him use it on his own arm to make a blade It’s kinda easily forgotten since the series is so fast, but alchemy at its core is science. Alchemists have to have a full understanding of what the materials they are transmuting are. Every time a transmutation is used there’s a lot of mental work being done under the surface Even a seemingly less intelligent character like scar has to fully understand the material he’s deconstructing. It’s why when he first fought Ed he didn’t destroy his automail arm with an attack. He didn’t realize Ed had an automail arm and was trying to deconstruct human flesh
