The actor who played Sam is actually deaf in real life, i love that they did that and Sam and Henry's story will always break me, the big infected is called a Bloater and i think, if i remember correctly, they have been 'alive' for a really long time, so the fungus just took over their whole body, inside and outside, but i'm not sure.
