December Q&A: DOI prohibition, 5-HT3 antagonists, prodrugs and more!
Added 2023-12-29 07:36:07 +0000 UTCIn this Q&A I discuss the DEA's new plans to prohibit DOI and DOC, Ritalin tablet prestidigitation, clandestine plant breeding and X-ray crystallography. A link to the Federal Register can be found below, enjoy!

2024-01-13 02:18:07 +0000 UTCYou can absolutely develop databases for rapid dereplication and identification of NMR data. I had to do this during postdoc. It was super tedious because the NMR data either 1- had to be entered manually or 2- freshly acquired (which isnt possible if you dont have all the compounda you want to out in your datase. Its called Spektraris and should be open source. But it works very well if you put the time into building a database of compounds you expect. Either way if you could have an AI read pdf files from scientific literature this would massively help to build a giant database. The issue there is AI wont be able to notice errors in literature. If you couls build such a large database you can probably start trainimg a AI tool to help with interpretation. Also yea Mestrenova kind of sucks. hah

2024-01-07 06:27:05 +0000 UTC4-aco-EPT and 4-ho-ept have a good bit more information available on them then EPT 4-aco-ept is mildly stimulating, lacking in a deep or confusing headspace. I find it useful for microdosing prior to social engagement or as an adjunct to other psychoactives Due to its versatile and mild character. Unlike many other psychedelics it can be administered alongside stimulants without inducing anxiety (for me) It is active at a 10-30mg dose for 1.5-2h.

2024-01-05 05:42:54 +0000 UTCthank you for answering my crystallography question! regarding machine learning for structure solutions, there's definitely software for single crystal XRD that have some structure solution functionality, but I don't know of anything similar for powder diffraction beyond basic peak matching with a data library as the density of defects is generally much higher in a polycrystalline materials and other pattern artifacts (i.e., peak broadening, preferred orientation, etc.). most crystallographers i know of (myself included) are hesitant to do full structure solutions from powder XRD without data collected using synchrotron radiation and/or other complimentary scattering methods like pair distribution function analysis or neutron diffraction, though I work in an exclusively inorganic materials space, so this may be different over in the organics world.

2024-01-02 11:16:16 +0000 UTCIt's where the peristalsis is your stomach slows. I have had it myself before, a paralytic illeus. And my grandmother calls it a lazy stomach because it's most likely easier than explaining the medical term to an everyday person who isn't interested in medical terms

2023-12-31 09:22:03 +0000 UTCJust curious, is there anyway of interviewing Leanord Pickard. I know you said in the past you that he doesnt like to talk about any of the chemistry he had done in the past. Would you know if he would be willing to talk about the case with skinner or his view on pyschedlics at his stage in his life? I think it would be an interesting synergy with you and him

2023-12-29 23:04:10 +0000 UTCHappy new year everyone! Hamilton, you mentioned the idea of using machine learning to interpret NMR spectra? Great idea for all sorts of spectroscopy, and they're on it! https://www.bruker.com/en/landingpages/bbio/artificial-intelligence-in-nmr.html. There's even a YT video called, "ChatGTP and NMR", and the applications to chemistry in general are just beginning to be tapped . From molecular modeling to alfafold, those darned stochastic parrots are gonna revolutionize things a little bit. I like the idea of incorporating biochemical steps to both simplify some synthetic pathways and create "green" alternativess. Maybe in the future we'll be trading "hot" yeast cultures instead of worrying about precursors. Is it OK to mention Sci Hub here? It's a great resource for papers over a year or so old. That probably goes for Chemical and Biological Abstracts as well, but up to date volumes should be available at university libraries ..very tedious though to search by hand! My generation grew up in the pre-digital era when digging through the immense collections of periodicals in the stacks of medical school libraries and photocopying articles of interest was a normal part of going to school. Now that almost everything is digitalized and available online it's a lot easier in some ways ....of course a five page article from the Journal of Organic Chemistry that one used to photocopy at the library for a dollar is now only available online and can cost $35.00 or more.....thus the beauty of Sci-Hub ....I feel that as long as it's for personal educational use only it's OK, but YMMV. Finally, even if the controlled substances laws were loosened it's going to be more difficult for people to explore medicinal chemistry at home than in the past in this post-Oaklahoma City, post-9/11 post fentanyl world.....and we're not even post-CRISPR yet! But hope springs eternal....Cheers ...

2023-12-29 20:32:55 +0000 UTCHi Xavier - I think so, why not? I'd imagine as more therapists use ketamine there'll be more reports of this, but there's already literature on using ondansetron for nausea during ketamine anesthesia, for example: https://www.sciencedirect.com/science/article/abs/pii/S0735675719305947. The results aren't 100% by any means, but it seems to be of some effect.

2023-12-29 19:34:38 +0000 UTCI feel dumb. Recently had my first big trip and elected not to take ondansetron because some person online said it might interfere with the experience. My trip sitter caught most of the barf in my designated barf pot, but was still left with a mess to clean up off the floor, while I just laid there, helpless and embarrassed in front of this cool guy I really liked and respected. Even after the stomach issues passed, I was still wracked and yelling "AUGH! AUGH!" because the nausea had somehow transcended my stomach, and yelling my head off seemed to be the only way to expel it. I thought, "what if the entire hours-long trip is like this?!" And then I thought, "well, sometimes things just have to suck for awhile." I realized most of my day-to-day mental energy was spent trying to avoid discomfort. Like, all I used to think about was a decision tree with bad outcomes and I would spend most of my time devising ways to avoid those bad outcomes. Basically, I had anxiety. But sitting through that torturous, bone-deep, full-body nausea just had me thinking: "Things have to suck sometimes. It's ok. Things have to suck sometimes. It's ok..." I took that to heart, and I just don't have anxiety anymore. But I'll take ondansetron next time.

2023-12-29 16:31:55 +0000 UTCIf it's a serious question, in vitro refers to biological studies performed outside of a living organism. Usually this means in a lab. It literally means "in glass", as in a test tube, flask, petrie dish, etc., as opposed to "in vivo" (in a living organism) or "in silico" (a computer simulation).

2023-12-29 10:28:34 +0000 UTCPublic comments were instrumental in keeping kratom unscheduled. More than 30,000 comments, many fron licensed medical professionals, completely surprised DEA and forced them to back down. They were trying to push it through under emergency scheduling and failed to meet most of the criteria to do so. Looks like they learned from that and have swung to blaming international treaty with WHO/UN instead butwhy?

2023-12-29 09:18:29 +0000 UTCThank you for answering my question! It’s quite odd to me that nobody has developed cultivars of most of the psychoactive plants besides cannabis. The only hypothesis I have is that it takes a long time to breed these plants and requires a lot of land to grow your trial fields which is difficult for most people. Psychoactive plants like coca and khat are woody perennial plants with long generation times in breeding programs, as well as being highly heterozygous outcrossers that don’t produce true to type seed and would require clonal propagation by cuttings/grafting. My guess is developing an exceptional cultivar with plants like these would take probably 10-20 years. I work in breeding/genetics research of woody perennial crops so if anyone is looking to try breeding psychoactive plants I’d love to help.
