Nooooooo, for example in this drawing i have everything in layers! The trashbin has its own lineart layer and beneath there is the colors in another layer! Then there are the characters with their color and then there are the rest of the room with a lineart layer and a color layer beneath! And for the outside is the same process! It is really easy to shade and add lightning if you work this way!
