Yes Jensen was married at the time as he married Danneel a few months after Jared married Gen, however, obviously Gen is known to the fans through her Ruby character whilst Danneel was not and I think having both wives would have taken too much time up. As has been said, on plenty of shows the leads don't get on, some even refuse to talk to each or interact with each other unless it's from the script on camera, hence they wanted to poke fun at those behind the scenes stories. Jared and Jensen have got on like brothers since they first met for the network interview. Most of the characters are real people or based on real people, although played by actors, I think the stunt coordinator was the only actual real person. They did break the 4th wall and both Jared & Jensen were keen to not play themselves but rather play Sam & Dean playing themselves. That isn't Jared's real house, although he has got that picture of him in the horse in his garage, and that isn't their real trailers, apparently much too flash, although they have played around with drones and toy helicopters so some aspects are true, others exaggerated. It's such a good episode.

Both Jensen and Jared got married in the same year (2010), a few months apart (Padaleckis at the end of Fenruary, and the Ackles' at mid-May - both weddings before season 6 even started being filmed) and both couples celebrated their 10 year anniversary this year. Around season 9 was when Jensen's first daughter was born. Danneel - Jensen's wife - didn't want to appear on the show, because she wasn't part of it like Gen was and at that point, she didn't feel like getting involved would be a smart thing to do. Gen making an appearance was part of the joke since she was known as Ruby to the viewers and it was funny to show Sam marring her.

Misha tried to make fun of himself, being the most shallow, self-obsessed version of himself he could be and was disappointed when a friend said how great it was to see him playing himself. Robert Singer did not name Bobby after himself. Bobby started out as just Bobby, but for a joke, the design staff made a sign for his junkyard with the name Singer and that became his name. Daneel was with Jensen at the time (not sure when they married), but hesitated to get involved with Supernatural. She knew how important it was to him and was afraid she would mess it up.

But there is a video from a SPN convention with her telling a story about traveling in Germany with Jared and a couple other people from the show on a train. It's really funny. To answer a couple of your questions...No Jensen was not married at this point in shooting. I believe it's not until somewhere around season 9 or 10 that he gets hitched. That's probably why they used his "trailer" instead of going to his house. The whole thing with 'at least they're talking' is a trope from a lot of older TV series over the decades. You hear stories years later about cast members secretly hating each other while filming. That's obviously not the case here. And I hear that Misha's actual Twitter account is a lot of fun to follow. I also wanted to give alternate Earth Vancouver PD props for letting two celebrity actors wander through an active murder scene and conduct their own interviews with witnesses. Very professional.
