As you saw, Dio can sense that the Joestar Bloodline has also unlocked stands thanks to him inhabiting Jonathan's body. Dio could go himself to fight them, but that is riskier to confront them directly when he doesn't know their powers. Alternatively, in Dio's eyes he prob sees them all as beneath him as well, not worthy to personally fight him.

Also people can correct me but I think the painting at the beginning is just a misdirect, I think he just shoots an emerald or cuts jojo's leg at the exact time he paints the red streak. And just happens to also be painting him. If that's wrong please correct me.
